fa اثر سد بر ساختار ژنتیکی گونه سیاه ماهی Capoeta capoeta gracilis در رودخانه تجن ژنتیک جانوری Subject 02 پژوهشی کامل Applicable <h1 dir="RTL"><span style="font-size:12px;"><span style="font-family:Tahoma;">سدها به طور بالقوه می‌توانند بر اندازه جمعیت و ارتباطات میان آن‌ها، کاهش تنوع ژنتیکی و افزایش تفاوت‌های ژنتیکی در میان جمعیت‌های ماهی رودخانه‌ای اثر بگذارند. بنابراین در این تحقیق، اثر سد شهید رجایی بر تنوع ژنتیکی سیاه ماهی در رودخانه تجن با استفاده از توالی‌های ژن سیتوکروم<span dir="LTR">b</span> &nbsp;مورد بررسی قرار گرفت. نمونه‌های سیاه ماهی از بالادست و پایین دست سد جمع‌آوری شدند. توالی یابی و تجزیه و تحلیل این ژن 11هاپلوتیپ را در نمونه‌ها نشان داد و تنوع نوکلئوتیدی و هاپلوتیپی به‌ترتیب 00062/0، 00939/0، 37778/0 و 70526/0 برای جمعیت‌های بالادست و پایین دست بود. تجزیه و تحلیل واریانس مولکولی (<span dir="LTR">AMOVA</span>)، 45/88 درصد از تغییرات ژنتیکی موجود در درون جمعیت و 21/6 درصد از تغییرات ژنتیکی در میان جمعیت‌ها را نشان داد. تجزیه و تحلیل پارامتر <em><span dir="LTR">F_ST</span></em> نشان داد که تفاوت ژنتیکی معنی‌داری بین جمعیت‌های مورد مطالعه وجود دارد. در مجموع می‌توان گفت تمایز ژنتیکی قابل ملاحظه‌ای در بین جمعیت‌های بالادست و پایین دست سیاه ماهی در رودخانه تجن مشاهده شد (05/0 = <em><span dir="LTR">P</span></em>). هر چند باید با احتیاط در مورد این نتیجه‌گیری صحبت کرد زیرا در این تحقیق فقط از یک ژن استفاده شده‌است. برای نتیجه‌گیری بهتر توصیه می‌شود از سایر ژن‌ها و هم‌چنین از سایر شاخص‌های ژنتیکی دیگر از قبیل ریز مارهواره‌ها استفاده شود و به علاوه مطالعات مشابه نیز در سایر مناطق انجام شود.</span></span></h1> Dams can potentially affect the size of populations and their connectivity, reduce genetic variation and increase genetic differences among isolated riverine fish populations. Therefore, in this research, the effect of Shahid Rajaee dam on the genetic variation of <em>Capoeta capoeta gracilis</em> in the Tajan River was investigated using sequences of cytochrome b gene. The <em>Capoeta capoeta gracilis</em> samples were collected from upstream and downstream of the dam. Sequence and analysis of this gene showed 11 haplotypes in the samples. Nucleotide diversity and haplotype diversity were 0.00062, 0.00939, 0.37778 and 0.70526 for upstream and downstream populations, respectively. Analysis of molecular variance (AMOVA) showed that 88.45% of genetic variation contained within population and 6.21% of genetic variation among groups. The analysis of the F_ST showed that there is a significant genetic difference between the studied populations. In total, it can be concluded that there was a significant genetic differentiation between the upstream and downstream populations of the <em>Capoeta capoeta gracilis</em> in the Tajan River (P = 0.05). However,&nbsp;this&nbsp;conclusion&nbsp;should&nbsp;be&nbsp;cautious, because only one gene was used in this research.
