fa بررسی بیان ژن SOX9 در بافت‌های تاسماهی ایرانی (Acipenser persicus) ژن SOX9 در میان مهره‌داران یکی از مهم‌ترین فاکتور‌های رونویسی جهت تکامل بسیاری از بافت-ها و ارگان‌ها و به‌طور ویژه تعیین جنسیت و غضروف‌زایی است. تاسماهی ایرانی (Acipenser persicus) یکی از گونه‌های مهم خانواده تاسماهیان بوده که بومی سواحل جنوبی دریای خزر می‌باشد. این گونه به دلیل ارزش اقتصادی آن و هم‌چنین بلوغ زودرس نسبت به دیگر گونه‌های پرورشی از جمله فیل‌ماهی، از گونه‌های موردنظر آبزی‌پروری در کشور می‌باشد. هدف از این تحقیق، شناسایی بخشی از توالی SOX9 و بررسی الگوی بیان آن در بافت‌های مختلف تاسماهی ایرانی با استفاده از تکنیک Real time PCR، بود. به این منظور ده بافت مختلف تاسماهی ایرانی جوان (آبشش، پیلوریک، طحال، گناد، کلیه، روده، قلب، پوست، کبد، ماهیچه) مورد مطالعه قرار گرفت. پس از توالی‌یابی قطعه‌ای به طول 210 نوکلئوتید شناسایی و سپس در بانک ژن NCBI (KP300013) ثبت شد. نتایج نشان داد که ژن SOX9 در اکثر بافت‌ها بیان شده و بیشترین میزان بیان (26/0 ± 5/0) در بافت آبشش مشاهده شد. با توجه به نتایج این تحقیق به نظر می‌رسد که در سنین پایین، احتمالا ژن SOX9 نقش کمتری در تعیین جنسیت داشته و بیشتر در فرآیند‌های تکاملی دیگر نظیر غضروف‌سازی دخالت دارد. T he SOX9 gene is one of the important transcription factors in the development of many tissues and organs, particularly in sex determination and chondrogenesis among vertebrates. Persian sturgeon (Acipenser persicus) is one of the most important species that is native to the southern coast of the Caspian Sea. This species due to economic value and early maturation compared to other farmed species such as Beluga is considered as main candidate species for aquaculture. The purpose of this study was to investigate the characterization of partial sequence of SOX9 and its expression in various tissues using Real Time PCR. Ten different tissues of Persian sturgeon (gills, pyloric, spleen, gonads, kidney, intestine, heart, skin, liver and muscle) were analyzed. Sequence analysis revealed a 210-bp cDNA and then it registered in NCBI gene bank (KP300013). The results showed that the SOX9 mRNA was detected in most tissues and the maximum expression was measured in gill (0.26±0.5).
