fa بررسی تنوع ژنتیکی ارقام و ژنوتیپ‌های امید بخش انگور وارد شده از کشور روسیه با استفاده از نشانگر DNA بین ریزماهواره‌ای(ISSR) Genetic Diversity of Promised Russian Grapevine Genotype and Varieties by Using Inter Single Sequence Repeat (ISSR) DNA Marker ژنتیک گیاهی Subject 01 پژوهشی کامل Applicable <div style="text-align: justify;">انگور یکی از محصولهای مهم باغی در دنیا و ایران به‌شمار میرود و از تنوع ژنتیکی بالایی برخوردار است. تعیین سطح تنوع ژنتیکی اولین قدم در اجرای برنامههای به‌نژادی است. اطلاع از تنوع ژنتیکی علاوه بر ارائه اطلاعاتی در زمینه روابط تکاملی و فیلوژنتیکی، نقشه ژنتیکی، سازگاری‌های اکولوژیکی و محیطی و در نهایت انتخاب والدین مطلوب در برنامه‌های اصلاحی، موجب کاهش مدت زمان و هزینه پروژههای اصلاحی خواهد شد. این تحقیق به‌منظور بررسی تنوع ژنتیکی 69 رقم و ژنوتیپ‌های امید بخش انگور روسی به‌همراه سه رقم انگور تجاری ایرانی با استفاده از 15 نشانگر بین ریزماهواره‌ای (ISSR) انجام گرفت. فراورده‌های حاصل از PCR بر روی ژل آگارز 3/1 درصد، الکتروفورز شدند. پس از اختصاص یک و صفر به وجود یا عدم وجود نوار در هر آغازگر برای هر ژنوتیپ، تجزیه خوشه‌ای به روش UPGMA بر مبنای ضریب تشابه جاکارد انجام گرفت. بیشترین درصد چندشکلی (100 درصد) مربوط به آغازگرهای UBC-823، UBC-825، UBC-841، UBC-846، UBC-855 و UBC-873 و کمترین آن نیز (7/66 درصد) مربوط به آغازگر UBC-817 بود. بیشترین و کمترین میران شاخص محتوای چند شکلی به‌ترتیب در آغازگرهای UBC-825 (487/0) و UBC-856 (216/0) مشاهده شد. بیشترین و کمترین میران شاخص نشانگر به‌ترتیب 718/2 و 904/0 به‌ترتیب مربوط به آغازگرهای UBC-889 و UBC-817 بود. از نظر شاخص EMR (Effective Multiplex Ratio) نیز بیشترین و کمترین مقدار به‌ترتیب 7 و 33/1 مربوط به آغازگرهایUBC-841 و UBC-817 بود. بیشترین و کمترین مقدار قدرت تفکیک به‌ترتیب 11/8 و 2 در آغازگرهای UBC-877 و UBC-841 دیده شد. دو آغازگر UBC-815 و UBC-834 نوارهای واضح و قابل امتیازی نداشتند. از 13 آغازگر نشانگر مولکولی ISSR، به‌طورکلی 73 نوار به‌دست آمد که 64 نوار چند شکل بودند و درصد چندشکلی کل 1/88% برآورد شد. بیشترین و کمترین تعداد نوارها به‌ترتیب هشت و دو نوار مربوط به آغازگرهای UBC-826 و UBC-817 بود. تجزیه خوشهای به روش UPGMA بر مبنای ضریب تشابه جاکارد منجر به طبقه‌بندی 72 رقم انگور با مقدار تشابه بین 89/0-12/0 در 7 گروه متفاوت شد. در مطالعه حاضر تنوع بالایی در میان ارقام انگور مورد مطالعه مشاهده شد. این امر ممکن است نشان‌دهنده متفاوت بودن منشاء جغرافیای ارقام و ژنوتیپهای مورد بررسی باشد.  </div> <div style="text-align: justify;">Grape is one of the most important horticultural products in the world and in Iran and has a high genetic diversity. Determining the level of genetic diversity is the first step in plant breeding programs. Knowledge of genetic diversity, in addition to providing information on evolutionary and phylogenetic relationships, genetic mapping, ecological and environmental adaptations, and ultimately selecting the right parents for breeding programs, will reduce the time and cost of breeding projects. This study was performed to investigate the genetic diversity of 69 cultivars and promising genotypes of Russian grapevines with three Iranian commercial grapevine cultivars using 15 microsatellite markers (ISSR). Polymerase chain reaction (PCR) samples were electrophoresed in 1.3% agarose gel. Cluster analysis with the UPGMA method was used based on the Jaccard coefficient after assigning one and zero to the presence or absence of a band in each primer for each genotype. The highest percentage of polymorphisms (100%) was related to UBC-823, UBC-825, UBC-841, UBC-846, UBC-855, and UBC-873 primers, and the lowest (66.7%) was related to UBC-817 primer. The highest and lowest levels of the polymorphic content index were observed in UBC-825 (0.487) and UBC-856 (0.216) primers, respectively. The highest and lowest levels of marker index were 2.718 and 0.904 related to UBC-889 and UBC-817 primers, respectively. The highest and lowest values of the effective multiplex ratio index were 7 and 1.33 for UBC-841 and UBC-817 primers, respectively. The highest and lowest resolving power were 8.11 and 2 for UBC-877 and UBC-841 primers, respectively. The UBC-815 and UBC-834 primers had no clear and concise bands. Overall, 73 bands were obtained from the 13 primers of the ISSR DNA marker which 64 bands were polymorphic and the total polymorphism was 88.1%. The highest and lowest numbers of bands were eight and two bands for UBC-826 and UBC-817 primers, respectively. The cluster analysis was performed by the UPGMA method based on Jaccard's coefficient of similarity, which led to the classification of 72 grape varieties with a similarity value of 0.12 to 0.89 in seven different groups. In the present study, high diversity was observed among the studied grapevine cultivars.
