fa بررسی تأثیر ضدسرطانی عصاره بذر زیره کرمانی بر بیان برخی ژنها در سلول‌های سرطانی MCF-7 ژنتیک انسانی Subject 03 پژوهشی کامل Applicable <h1 dir="RTL" style="text-align:justify"><span style="font-family:Tahoma;"><span style="font-size:9pt"><span style="direction:rtl"><span style="unicode-bidi:embed"><span class="Heading1Char"><span style="font-weight:bold"><span lang="AR-SA" style="font-size:10.0pt">زیره کرمانی یکی از مهم‌ترین گیاهان دارویی بومی ایران است که دانه‌های آن در رژیم غذایی استفاده می‌شود و اثرات مضر جانبی برای انسان ندارد. مطالعات گذشته تأثیر دارویی این گیاه را در زمینه‌های مختلف نشان داده است اما تاکنون مطالعه‌ای در مورد تأثیر ضدسرطانی بذر این گیاه صورت نگرفته است. در این مطالعه، اثر ضد سرطانی عصاره دانه زیره کرمانی بر سلول‌های سرطانی رده پستان </span><span dir="LTR" lang="X-NONE" style="font-size:10.0pt">MCF-7</span></span></span> <span class="Heading1Char"><span style="font-weight:bold"><span lang="AR-SA" style="font-size:10.0pt">موردبررسی قرارگرفته است. در ابتدا با آنالیز متن‌کاوی مسیرها و ژن‌های مهم متأثر از عوامل شیمی‌جلوگیری کننده گیاهی در درمان سرطان مشخص شد. بعد از عصاره‌گیری از بذرهای زیره کرمانی، ابتدا با استفاده از روش </span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span dir="LTR" lang="X-NONE" style="font-size:10.0pt">Neutral Red</span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span lang="AR-SA" style="font-size:10.0pt"> میزان زنده‌مانی سلول‌های </span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span dir="LTR" lang="X-NONE" style="font-size:10.0pt">MCF-7</span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span lang="AR-SA" style="font-size:10.0pt"> تحت تیمار با غلظت‌های متفاوت عصاره مورد بررسی قرار گرفت. سپس برای بررسی تأثیر عصاره بر فرایند آپوپتوز از تکنیک فلوسایتومتری استفاده شد. در آخر بیان چند ژن‌ مهم به‌دست‌آمده از آنالیز متن‌کاوی با استفاده از روش </span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span dir="LTR" lang="X-NONE" style="font-size:10.0pt">real-time PCR</span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span lang="AR-SA" style="font-size:10.0pt"> ارزیابی شد. تأثیر عصاره بر میزان زنده‌مانی سلولی به‌صورت وابسته به غلظت بود. عصاره بذر زیره در دوز 160 میکروگرم بر میلی‌لیتر باعث کاهش زنده‌مانی سلولی به میزان 50 درصد در سلول‌های </span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span dir="LTR" lang="X-NONE" style="font-size:10.0pt">MCF-7</span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span lang="AR-SA" style="font-size:10.0pt"> شد. تحت تیمار با عصاره زیره ژن‌های به‌دست‌آمده از آنالیز متن‌کاوی به نحوی تغییر نشان دادند که می‌تواند در درمان سرطان تأثیر مثبتی داشته باشد. مطالعه حاضر برای اولین بار نشان می‌دهد که عصاره بذر زیره کرمانی می‌تواند آپوپتوز را در سلول‌های سرطانی </span></span></span><span class="Heading1Char"><span style="font-weight:bold"><span dir="LTR" lang="X-NONE" style="font-size:10.0pt">MCF-7</span><span lang="AR-SA" style="font-size:10.0pt"> القا کند. </span></span></span></span></span></span></span></h1> <span style="font-size:11pt"><span style="line-height:150%"><span calibri="" style="font-family:"><span style="font-size:12.0pt"><span style="line-height:150%"><span new="" roman="" style="font-family:" times="">Black cumin is one of the most important native medicinal plants of Iran, which the seeds of this plant are used in the diet and have no side effects for human. Previous studies have shown the medicinal effect of this plant in various fields, but so far, no study has been done on the anti-cancer effect of the seeds of this plant. In this study, the anti-cancer effect of cumin seed extract on MCF-7 breast cancer cells was investigated. Firstly, the analysis of important pathways and genes affected by phytochemical inhibitors in the treatment of cancer was identified by text-mining. After extracting cumin seeds, the viability of MCF-7 cells treated with different concentrations of the extract were investigated using the Neutral Red method. Then flowcytometry technique was used to evaluate the effect of the extract on the apoptotic process. Finally, the expression of several important genes obtained from text-mining analysis was evaluated using real-time PCR. The effect of the extract on cell viability was dependent on the concentration. Black cumin seed extract at a dose of 160 &mu;g/ml reduced cell viability by 50% in MCF-7 cells. Treated with black cumin extract, the genes obtained from text-mining analysis showed a change that could have a positive effect on cancer treatment.
