Tomato yellow leaf curl viral disease (TYLCV) is one of the main limiting factors that has caused great damage to tomato crop in tropical and subtropical regions of the world The present study was carried out to evaluate the role of transmission and pyramiding of genes (TY-2, TY-1/3), effective in creating resistance to this viral disease in several different tomato lines (sensitive but optimal performance). For this purpose, after crossing resistant parent to the desired lines, three steps back-cross and a self-fertilization step were performed, and then several traits involved in the plant's defense system, also the concentration of the genome of TYLCV virus, were examined using polymerase chain reaction technique to evaluate resistance. The results of this study showed that viral contamination increased the level of phenol and flavonoid compounds also antioxidant enzymes in the control plant. While lines that carried genes TY-2 and TY-1/3 had a lower level of synthesis and expression of non-enzymatic antioxidant compounds (phenol and flavonoid) and antioxidant enzymes (catalase, peroxidase, guaiacol peroxidase, and polyphenol Oxidase). Also, in the survey of the concentration of the genome of the TYLCV virus , it was observed that lines containing two gene loci Ty-2 and TY-1/3 had the lowest level of virus genome than in the control plant. These results indicated low levels of virus proliferation and resistance as the result of the presence of the two genetic loci in the host plant. According to the observations of this study, by integrating different TY genes desired levels of resistance in desirable sensitive lines against TYLCV virus, can be obtained.
