In order to study the proteome response of chickpea genotypes under drought stress, two tolerant and sensitive genotypes were treated to stress and non stress in greenhouse conditions. After protein extraction and drawing a standard curve, the first dimension electrophoresis leads to separate of proteins in terms of electrical focusing and the second dimension electrophoresis separates the proteins in terms of molecular weight. The results of the imaging and analysis of gels related to two tolerant genotypes of Karaj-3628 and sensitive genotype Fasa-6880 under two conditions of stress and nonstress showed that there were significant differences among the 401 repeatable spots. The tolerant genotype of Karaj-3628 in nonstress and stress conditions, 11 and 11 spots increased expression and 13 and 10 spots decreased expressions, and also sensitive genotype Fasa-6880 in nonstress and stress conditions, 16 and 7 spots increased expression and 7 and 18 spots decreased expressions respectively. The GAPDH type A increased expression in tolerant genotypes and decrease expression in sensitive genotypes, The Thioredoxin h1 at different levels of stress increased expression, The EST mhl-0016-B10 in stress treatment decreased expressions, The small subunit of Robisco's enzyme increased expression under stress, The 20S proteasome subunit alpha-1 increased expression under stress and decreased expression in sensitive genotypes,The Putative cytochrome c oxidase subunti 6b-1 decreased expression under stress treatment in both tolerant and sensitive genotypes. Comparison of amount of volume percent by inter class report methods indicated of expression increasment of superoxide dismutase in gel of two genotypes under stress.
