fa کلونینگ و تعیین توالی نوکلئوتیدی ژن کامل پروتئین ماتریکس ویروس بیماری نیوکاسل شایع در گله‌های مرغ گوشتی شمال شرق ایران Cloning and nucleotide sequencing of the complete matrix protein of Newcastle disease virus subgenotype VII.1.1 prevalence in broiler flocks of northeastern Iran ژنتیک جانوری Subject 02 پژوهشی کامل Applicable <h1 style="text-align: justify;">بیماری نیوکاسل یک بیماری ویروسی شناخته شده و یکی از اصلی‌ترین عوامل بیماری‌زا در صنعت طیور در سراسر جهان است. پروتئین ماتریکس (M) یکی از پروتئین‌های ساختاری ویروس بیماری نیوکاسل است که نقش مهمی در بیماری‌زایی، تکثیر و جوانه‌زنی ویروس دارد و همچنین پوشش ویروسی را به کمپلکس ریبونوکلئوپروتئین (RNP) پیوند می‌دهد. با توجه به نقش پروتئین ماتریکس، این مطالعه با هدف فراهم نمودن اطلاعات تکمیلی جهت تعیین هویت یک جدایه ویروس بیماری نیوکاسل با نام CH/RT40/IR/2011 شایع در گله‌های مرغ گوشتی تجاری استان خراسان رضوی انجام گرفت. ژن کامل پروتئین ماتریکس RT40 با استفاده از پرایمرهای اختصاصی تکثیر و سپس در پلاسمید PTZ57r/t کلون شد. پس از تعیین توالی نوکلئوتیدی تجزیه و تحلیل داده‌ها با استفاده از نرم‌افزار CLC Main Workbench 5.5 انجام شد. درخت فیلوژنتیک با استفاده از روش Maximum likelihood با نرم‌‌افزار MEGA6 رسم شد. تعیین توالی ژن پروتئین ماتریکس (1241نوکلئوتید)، درخت فیلوژنتیک و آنالیز فواصل ژنتیکی بین ژنوتیپی نشان داد که  RT40با تحت ژنوتیپ VII.1.1 ویروس‌های بیماری نیوکاسل ایرانی (MF417546) و عراقی (MT370497,MT370499) شباهت دارد. بررسی توالی پروتئینی سیگنال‌های متمرکزکننده هسته‌ای (247KKGKKVTFDKIEEKIRR263) و دیگر نواحی پروتئین ماتریکس RT40 نشان داد که این ویروس بسیار شبیه به ویروس‌های فوق حاد بیماری نیوکاسل است. یافته‌های حاصل از این مطالعه تکمیل‌کننده اطلاعات لازم برای شناسایی کامل جدایه RT40 و معرفی آن به‌عنوان یک سویه بومی ویروس بیماری نیوکاسل بود.  </h1> Newcastle disease (ND) is a well-known and major viral disease of poultry industry all over the world. Viral matrix (M) protein is a structural protein linking the viral envelope with the virus core that plays an important role in pathogenesis. It plays a crucial role in virus assembly and interact with the ribonucleoprotein (RNP) complex as well as with the viral membrane. Due to the role of matrix protein, this study was performed to complete previous studies to identify a Newcastle disease virus (NDV) isolate naming CH/RT40/IR/2011 which is prevalent in commercial broiler flocks in Khorasan Razavi province. The full M protein gene of the RT40 was amplified using specific primers and then cloned into a PTZ57R/T cloning vector. The cloning vector product was sequenced from both directions and data analyses were performed using CLC Main Workbench 5.5. The phylogenetic tree was constructed with the maximum likelihood method in MEGA 6. Nucleotide sequencing of M protein gene (nt: 1241), phylogenetic tree and analysis of genetic distances between genotypes showed that RT40 was similar to Iranian (MF417546) and Iraqi (MT370499, MT370497) NDVs subgenotype VII.1.1. Investigation of amino acid sequence of the nuclear localization signals (247KKGKKVTFDKIEEKIRR263) and other matrix gene protein regions revealed that this virus was very similar to very virulent NDVs.
